چکیده

Several cDNA clones of the protozoa Trypanosoma brucei and Trypanosoma lewisi were isolated as a reult of selection for evolutionarily conserved genes encoding cell-surface proteins (Lee and Van der Ploeg, unpublished). cDNA clone Tb-17 had a predicted open reading frame, encoding a 23 kDa protein (Figure 1A). This protein shared significant homology with a flagellar Ca binding protein (FCaBP) previously identified in Trypanosoma cruzi (1, Figure IB). The predicted Tb-17 protein encoded the 'E-F hand' Ca binding site homologous to the Ca binding site of Calmodulin in T. brucei (2, 3). Tb-17 is expressed at a relatively high level in both the bloodstream form and the insect form of T. brucei (data not shown). The evolutionary conservation, the presumed flagellar location and its potential Ca binding ability, suggest that this protein may play a role in cell motility or in Ca transport.

A novel flagellar Ca2+-binding protein in trypanosomes.

A 24-kDa protein of Trypanosoma cruzi, the protozoan parasite that causes Chagas' disease, is recognized by antisera from both humans and experimental animals infected with this organism. Near its C terminus are two regions that have sequence similarity with several Ca2+-binding proteins and that conform to the "E-F hand" Ca2+-binding structure. We expressed a cDNA encoding this protein in Esch...

Extensive flagellar remodeling during the complex life cycle of Paratrypanosoma, an early-branching trypanosomatid.

Paratrypanosoma confusum is a monoxenous kinetoplastid flagellate that constitutes the most basal branch of the highly diverse parasitic trypanosomatids, which include human pathogens Trypanosoma and Leishmania This makes Paratrypanosoma uniquely informative for the evolution of obligatory parasitism from free-living lifestyle and the evolution of human parasitism in some trypanosomatid lineage...

The Limits on Trypanosomatid Morphological Diversity

Cell shape is one, often overlooked, way in which protozoan parasites have adapted to a variety of host and vector environments and directional transmissions between these environments. Consequently, different parasite life cycle stages have characteristic morphologies. Trypanosomatid parasites are an excellent example of this in which large morphological variations between species and life cyc...

Flagellar and ciliary beating in trypanosome motility.

The single flagellum of Leishmania and Trypanosoma parasites is becoming an increasingly attractive model for the analysis of flagellar function-driven largely by the abundance of genomic and proteomic information available for the organelle, the genetic manipulability of the organisms and the importance of motility for the parasite lifecycle. However, as yet, there is a paucity of published da...

Cytoskeletal Association Is Important for Differential Targeting of Glucose Transporter Isoforms in Leishmania

The major glucose transporter of the parasitic protozoan Leishmania enriettii exists in two isoforms, one of which (iso-1) localizes to the flagellar membrane, while the other (iso-2) localizes to the plasma membrane of the cell body, the pellicular membrane. These two isoforms differ only in their cytosolic NH2-terminal domains. Using immunoblots and immunofluorescence microscopy of detergent-...
